Sam and Dean have a pretty damn complicated relationship, founded on a basically unhealthy life, extremely unfortunate events, and generalized chaos; as a child, Sam looked up to Dean quite a bit, though he'd slowly started realizing that not only was this life something to walk away from, but his brother wasn't always the best lead to follow — he didn't want to be a hunter, and he didn't want to follow his father. So Sam left for a plethora of reasons, and for the years he'd left he genuinely thought his family didn't want him back. When Dean finally reappeared, Sam had harbored a lot of negative feelings over the last fight that went down.
However, over the course of season 1, Sam rekindles the brotherly love that had drifted a bit since then. He tells Dean he would die for him, and he means it — and in a lot of ways, Sam is still very reliant on his brother's word and care. It's how he's always been: he never depended on John, but on Dean, and in that sense, Dean has been his most important 'parent'. Of course they have spats like siblings just fine. The combination of caregiver and annoying big brother makes him easier to lose his patience, rebel, be younger all over again, but ultimately Dean's his only current constant from his point in canon. And dammit, he loves the guy.
Since arriving on the Tranquility, Sam's been mostly relieved to find Dean alive and in one piece, but at the same time, he's already a little freaked at how secretive his brother is being. What would make him like that? What did Sam do, to really be locked out from learning the truth back home? Maybe he should just drop it, but when it comes to Dean, not knowing what the other knows is practically driving him up the walls. Knowing his brother is keeping things from him... it doesn't sit well, and Sammy's already a nervous type to begin with.
Jo's made a pretty damn good impression on him — courteous, kind, but also there's an obvious spark to her that makes it obvious how and why she ends up friends with Winchesters. She's pretty tight-lipped about the future like Dean, but her confidence and reassurance help to ease the dark clouds whirling in his head made a great impression. And even better, she offered a place and her time for when Sam really needed it. He's glad to know he has some people he can trust. And he'd be more than happy to forge that friendship all over again.
Also, please don't tell Dean he cleaned the bar like a determined housemaid.
